The 49^th TCS was formed in July 1942 from the redesignation of the 49^th Transport Squadron constituted a month earlier. It belonged to the 313^th Troop Carrier Group during all of its existence, and its history matched that of the group (which see). Aircraft of the 49^th TCS wore designation H2 on the nose.
